心筋の増殖を制御する機構を明らかにし,傷ついた心臓の再生を目指す
哺乳類の成体の心臓には、心筋梗塞などで障害を受けた後の再生能力がありません。その主因として、哺乳類の成体の心臓のポンプ機能を担う心筋細胞には、ごく一部のものを除いて、細胞分裂によって増殖する能力がないことが挙げられます。しかし同じ哺乳類でも、胎児期や生後すぐの新生児期には多くの心筋細胞が増殖能を保っており、心臓には再生能力があることが分かっています。我々はこれまでに、ミトコンドリア好気呼吸とそれによって生じる酸化ストレスが生後の心筋細胞の増殖能を失わせる原因の一つであることを見出しました。そこで、我々の研究室では、酸化ストレスにより細胞周期が制御されるメカニズムをより詳細に明らかにし、その機構をターゲットにした新しい心臓再生の方法を確立することを目指します。より具体的には、一部の心筋細胞が生後も増殖能を保つ機構、そしてミトコンドリア好気呼吸の低下による心筋細胞の増殖誘導の機構を理解し、そのエンジニアリングによる心筋再生の誘導の可能性を探ります。

Heart disease is the number one killer of humankind mainly because the heart does not have regenerative capabilities. Our previous study indicated that oxidative stress is the main inducer of cell cycle arrest in postnatal mammalian heart muscle cells, cardiomyocytes. Our research interest is focused on why postnatal mammalian cardiomyocytes do not proliferate after injury- and age-related myocardial damage, and whether it is possible to re-awaken endogenous regenerative capacity. We utilize molecular and cellular tools and mouse genetics to understand the role of oxidative metabolism in cardiomyocyte cell cycle regulation throughout the life cycle of mammals.
